Penetration pathway of metallic nanoparticles in normal and abnormal mouse skin

Abstract
As usefulness of drug delivery via skin is revealed, various methods to increase penetration efficiency are studied. But, it is very difficult that is penetration of drug into the skin without metamorphosing skin barrier. We studied possibility of permeation to stratum corneum and penetration pathway using metallic nanoparticles. This experiment compared permeation of metallic nanoparticles using confocal microscope and transmission electron microscope in normal
and abnormal mouse skin. Normal skin group observed that most nanoparticles permeated to intercellular lipid layer between corneocyte of outer stratum corneum. Also, confirmed that nanoparticles permeate to hair follicle and it is flowed in pilosebaceous duct. But, it couldn¡¯t observe that nanoparticles permeated to the other cells on around hair follicle. In contrast, we observed that nanoparticles permeated to lower stratum corneum in abnormal skin group. We are confirmed that if don¡¯t induce physical or chemical distortion to stratum corneum then penetration via stratum corneum is very difficult in nanoparticle¡¯s case. Also, confirmed that most nanoparticles permeated to hair follicle, but it is thought that additional research may need how to penetration into skin. This research expects to become basis of transdermal drug
transport pathway.
